AMYn1 aptamer (Uracil replacing Uad)

This sequence was inspired from a research paper published in 2017 by Minagawa et. al. The paper described an enzymatically synthesized aptamer targeting alpha-amylase called AMYm1. This aptamer contained (E)-5-(2-(N-(2-(N6-adeninyl)ethyl))carbamylvinyl)-uracil, or Uad, in place of thymine. AMYm1 binds to sAA with extremely high affinity (Kd < 1 nM).

This part is a modified version of the AMYm1 aptamer, where Uad is replaced with Uracil. This modification prevents the aptamer from binding specifically to alpha-amylase. In experiments that test experimental aptamers on alpha-amylase, this part can serve as a negative control to determine non-binding to alpha-amylase.

As of October of 2023, iGEM does not allow teams to upload RNA parts that contain uracil. As this part contains uracil, the correct RNA sequence is: GGATACCTTAACGCCGCCTATTGUGAACGACGUGAAUAGUGUUUGUGGGUCCGGAGUUGCACCCGUCUCGAAAUC.
