AD1.S5 - STAR Sense from pAD1 Plasmid Attenuation System



AD1.S5 is one of the sequences from the pAD1 plasmid attenuation system that produce Small Transcription Activating RNA (STAR) sense RNA. It and its corresponding sequence, the AD1.A5 (BBa_K4850001), from the two different plasmids can be used as a switch that control the on/off state of the transcription. STAR sense RNA possess an intrinsic terminator. In the absence of the STAR antisense RNA, the STAR sense RNA the intrinsic terminator would be formed, inhibits the transcription from happening. On the other hand, the intrinsic terminator would be sequestered when the STAR antisense RNA present, allowing the initiation of the downstream transcription.


Usage and Biology

The NUS-Singapore 2023 iGEM team constructed a plasmid contains both AD1.A5 and AD1.S5, called “STAR 1-Plasmid System”, facilitating the data generation process supported by their SIGNAL Assessment Workflow to generate sufficient quality data to train their model “OTTER”.
