A single gene for the fusion of three functional domains of cellulase (BG,CBH,EG) was designed by fusion strategy.The fusion gene can degrade cellulose into monosaccharides.Primers are designed to fuse the catalytic domains of these three enzymes . The reverse coding sequence of the flexible joint (G4S)3 (sequence GGGGSGGGGSGGGGS) was added to the reverse primers BG-L-XBAI-R and CBH-L-ECORI-R of BG and CBH. Two segments (G4S)3 serve as linking chains to the CD domain of the three cellulases. At the same time, a suitable restriction enzyme site was added to the end of the primer to facilitate the fusion of DNA and cloned genes.
