NarK promoter fused to sfGFP
narK promoter driving superfolder GFP expression. The part should promote expression of GFP under low oxygen conditions.
Testing indicates this does respond very well to oxygen. It shows no GFP fluorescence anaerobically or aerobically but gives GFP fluorescence at 0.5% oxygen and 2% oxygen.
This part is expected to only promote expression under low oxygen conditions. The composite part was grown in E. coli under 4 different oxygen conditions. Anaerobic, 0.5 % oxygen, 2% oxygen and fully aerobic (18% oxygen).
The graph shows how this promoter (Promoter B in the image) responded with 2 biological and 3 technical replicates per measurement point.
As can be seen this promoter responds to oxygen such that expression of superfolder GFP occurs at oxygen concentrations of 0.5% and 2% but not either fully anaerobically or under fully aerobic growth conditions.
