Since the tetR gene can repress the pTet promoter (BBa_R0040), the downstream genes of pTet, such as the cry gene and the GFP gene(BBa_E0040), can also be repressed.
While the switching temperature is achieved in the incubator, pTet as well as its downstream genes will be switched off. In this condition, almost none of the green fluorescent protein can be detected.
However, pTet and its downstream genes will not be switched off below the switching temperature.
In other words, we can observe a great amount of the green fluorescent protein only in sub 37°C environment.
