gRNA for Cystic Fibrosis Wildtype gene targeting of Cas12a / Cpf1


This part encodes for a guide RNA that can be used with Cpf1. The sequence includes a crRNA repeat, known as the fixed loop, to allow Lab Cas12a to recognize the gRNA (different manufacturers may produce Cas12a that are optimized for recognizing different fixed loops; the sequence posted here is optimized for NEB Cas12a). The sequence also includes a segment that is specific to the wild type CFTR gene. This part may be used to simply manufacture this gRNA. It can be used in conjunction with template DNA extracted from patients or synthesized in a lab to allow Cas12a to recognize and bind to the wild type CFTR gene. This may then be paired with further downstream effects (such as Cas12a activity causing fluorescence of some kind) to detect DNA samples that do not have the cystic fibrosis disease. This part is also useful as a control for use in conjunction with BBa_K2961004
