Bacterioferritin (BFR) with sfGFP conjugation
Bacterioferritin is a storage protein that can store iron ions so that the host cell becomes responsive to magnetic field. Conjugation with sfGFP reporter gene is provided by GS linker.
Usage and Biology
Bacterioferritin is one of the iron storage protein in bacteria. It forms hollow shell from 24 identical subunits, incorporating 12 haem groups. It has ferroxidase center that binds Fe2+ ions, oxidizes them by dioxygen to Fe3+, and participates in the subsequent Fe3+ oxide mineral core formation within the central cavity of the protein complex.
Bacterioferritin has a heme group normally, which is involved in release of iron to reduce immobilized Fe3+ to more soluble Fe2+. Therefore, we use the BFR gene which is provided by iGEM14_Berlin Team. It has heme group mutation that allows to keep iron ion inside the BFR protein to maintain magnetic property of the bacteria.
GS Linker with sfGFP is added to BFR to see expression of the BFR protein under the fluorescence microscope.
