As per BBa_K180002, omitting the RBS and STOP biobrick parts (B0034 and B0015) for speed and ease of construction.
The lacIq sequence preceding the tac promoter ensures that tac pR is in the 'OFF' state naturally. Green fluorescent protein is produced if IPTG successfully induces the promoter, demonstrating the regulatory effect of Lac upon the promoter.
