Hepatitis Delta Virus(HDV) Ribozyme

The hepatitis delta virus ribozyme is a self cleaving ribozyme originating from hepatitis delta virus. The cleavage occurs at the ribozymes 5' end independent from the upstream sequence. Therefore it is highly suitable to create defined ends for functional RNA expression. Furthermore its behvior is specific and shows good efficiency.

Therefore this ribozyme is used for all BBF RFC 110 parts derived from BBa_K1614002, as the 3' limiter. Thereby the production of RNA product with the user-defined ends is ensured. For more details have a look at File:BBF RFC 110.pdf

Fig.1: PAGE analysis of HDV cleavage behavior. The RNA of interest appears as a defined band under UV-shadowing. Higher yields for the desired product with high fidelity can be obtained by the PCR extraction of the transcription cassette with BBa_K1614005 BBa_K1614006. Uncleaved product can be observed as the third band.
