Usage and Biology

The Effects on tyrosine metabolism of csrA

CsrA is known as carbon-storage regulator protein, which regulates the expression of enzyme genes involved in glycolysis. That regulation process is based on the binding of CsrA and the mRNAs of enzymes. It was shown that csrA repression leaded to increase of the tyrosine production in previous study.

RNA Silencing Utilizing MicC sRNA Scaffold

The combination of the target-binding sequence of a gene to be repressed with MicC sRNA scaffold(Part:BBa_K1124005) works as a tool of RNA silencing (see descriptions in MicC sRNA scaffold(Part:BBa_K1124005)). In this part, the target-binding sequence is complementary to the first 24bp of csrA CDS(that is, the sequence from start codon ATG and the following 7 triplet codons). We expect that this part will repress csrA expression in post-transcriptional way and therefore will increase tyrosine production. Combining this part with anti-tyrR sRNA(Part:BBa_K1124009), we constructed tyrosine synthesis device(Part:BBa_K1124106), which can likely increase tyrosine production.
