LuxR+3OC6HSL independent R0065
This part is derived from R0065, a hybrid LuxR+3OC6HSL activated, CI repressed promoter. To eliminate LuxR+3OC6HSL dependence, the weak -35 hexamer of R0065 was changed to the consensus TTGACA sequence. Then, to increase the consitutive transcription rate, two modifications were made. First, a single base was been added to the region between the -35 and -10 elements, bringing that region to the optimal length of 17bp. Second, an 'extended -10' sequence was added 1 base pair upstream of the -10 hexamer. The two CI operators OR1 and OR2 were left unchanged. Taken together, this should be a strong CI-repressible promoter.
