Examining the effect of a single base mutation in the sequence prior to RBS on RFP production

We are testing the effect of changing the 5th base pair upstream of a known RBS sequence from A to G on the amount of RFP expression. The relative red fluorescence protein (RFP) data indicates the translation efficiency of mRNA. By changing this single base pair, the relative RFP expression will be lesser, suggesting that the translation efficiency decreases when the RBS sequence is mutated.
