An aconite isomerase from Bacillus thuringiensis.

This is an aconite isomerase from Bacillus thuringiensis YBT-1520, which can catalyze the isomerization of cis-aconite to trans-aconite and its reverse reaction at the same time, but has a higher activity to catalyze the isomerization of cis-aconite to trans-aconite.

Usage and Biology

TAA biosysnthesis-related gene A (tbrA) encodes a aconite isomerase in Bacillus thuringiensis YBT-1520, which can isomerize cis-aconite into rice aconite, and also has the ability to catalyze its reverse reaction. This property allows bacteria with such enzymes to isomerize TAA into CAA through aconite isomerase, thus growing on the medium with TAA as the only carbon source.[1] In our project, we aim to utilize the nematocidal activity of TAA as an alternative approach for killing nematodes. Additionally, we use the tbrAB genes, which encode an aconitate isomerase and a transport carrier, respectively. These genes allow the transport of TAA produced inside the bacteria to the extracellular environment, enabling our engineered bacteria to secrete TAA and thus exert a nematocidal effect.
