Strep-DigA-Long Linker-Fok_i

This part unites some of our basic parts. The combination of a StrepTag, an Anticalin-Tag and the linked protein domain Fok_i makes it possible to purify and detect the function of the protein. The DigoxigeninA tag allows the coupling to a digoxigenin linked oligonucleotide.


Usage and Biology

This composite part is one part of our universal endonuclease and needs another composite part like BBa_K243024 to build a functional heterodimer. The DigA tag leads the part to DNA which is hybridized with a Digoxigenin labeled oligonucleotide. The long linker creates a distance of 36bp between the DigA and the linked Fok_i protein domain. The StrepTag serves as a purification tag for Streptavidin column purification.
