Cas9 handle-reverse repeating sequence in gRNA
The Cas9 handle, specifically the stem-loop sequence of the guide RNA (gRNA), refers to the palindromic sequence within its structure. This sequence is capable of folding back upon itself to form a secondary structure, namely a stem-loop configuration, due to the hydrogen bonding between complementary nucleotide bases. Within the gRNA, this conserved sequence facilitates the stabilization of the gRNA-Cas9 protein complex.
