Cas9 handle-reverse repeating sequence in gRNA

The stem-loop sequence of gRNA (guide RNA) refers to the reverse repeating sequence in its structure, which can be folded back to form a secondary structure, that is, a stem-loop structure, due to the hydrogen bond pairing between complementary bases. In gRNA, this conserved sequence helps stabilize gRNA binding to Cas9 protein.
