Design Notes

Since we expect that this part will be used in the context of a fusion protein, we have modified this protein to be consistent with BBF RFC-12 Standard. We have submitted this part to the part registry as part BBa_K265002. Sequence taken from first 37 amino acids in phaZ1 gene. The signal peptide cleavage site is predicted between Ala37 and Leu38 therefore target protein will get cleaved off if fused after Ala37. Target proteins were seen to make a halo of translucence around cells on an opaque background denoting secretion of target protein.
