Design Notes

This part was designed with the same structure of the Berkley constitutive promoters. Indeed, the operator sequence was digested with SpeI and PstI and the same was done for the RFP. The two parts were then ligated. As a second passage, the obtained O2-RFP part was digested with XbaI and PstI and ligated downstream of the BBa_J23118 promoter digested with SpeI and PstI.This was done to allow an easier cloning of the operator and promoter sequences that are short in length (about 20 bp and 35 bp, respectively).
