spider silk protein MaSp1
MaSp1 encodes for spider silk, which is a highly repetitive amino acid sequence and has the properties of being strong, flexible, and elastic. This amino acid sequence is one of the shortest compared to other amino acid sequences for spider silk proteins listed in the paper by Tian et al, 2003.
MaSp1 amino acid sequence had several repeat amino acid sequences. After converting them into DNA sequences, our team designed the primer set to amplify MaSp1 via the PCR technique. However, the primer set kept annealing to several places on the DNA sequences via the Snapgene app. After consulting with the company, they have suggested our team use 3 different nucleotide codons for the same amino acid to make the DNA sequence not repeat much. Our team also added the start codon, ATG, at the 5'end of the MaSp1 DNA sequence so the RNA transcription would start from ATG on MaSp1 DNA
