Design Notes

sgRNA is a synthetic fusion of the endogenous bacterial crRNA and tracrRNA that provides both targeting specificity and scaffolding/binding ability for Cas9 nuclease. In order to construct the IbGBSSI knockout system, the appropriate sgRNA targeting for the gene IbGBSSI (BBa_K4887001)should be designed and ligated to the plasmid psgR-Cas9-At.

It was selected using the online sgRNA design tools and the available I. batatas draft genome sequences (Yang et al., 2017).
