Part:BBa_K4885007
1
This part includes a NGCatcher sequence BBa_K4119013 and bcd gene (BBa_K4119027) derived from Clostridium tyrobutyricum (C. tyrobutyricum) L319. It codes a fusion protein of NGCatcher and butyryl-CoA dehydrogenase (bcd). bcd gene codes for butyryl-CoA dehydrogenase which converts crotonyl-CoA to butyryl-CoA in the butyrate synthesis pathway in C. tyrobutyricum. NGTag/NGCatcher peptides are variants of the SpyTag/SpyCatcher system and are reported to be useful for peptide-peptide interaction for multienzyme assembly. NGCatcher has a high density of negative charge on its surface and is a highly charged protein. It forms a covalent bond through an isopeptide bond with NGTag rapidly which is not affected by the change of environment. The fusion protein of NGCatcher and butyryl-CoA dehydrogenase can be assembled with another enzyme fused with NGTag.
