PchA

The pchA is present in the butyrate sensor derived from the EHEC strain O157: H7, which was discovered by Nakanishi et al. (2009). The butyrate sensing system includes sequences of pchA promoters(PpchA) and pchA regulators located upstream, and LEE1 promoters(PLee1) and RBS located downstream. Nakanishi et al. found that leucine response regulatory protein (Lrp) forms a complex with butyrate, which activates the transcription of pchA regulatory factors after binding to the PpchA. The pchA regulator then binds to the PLee1 to transcribe the ler gene. In short, it is a regulatory gene(regulates LEE1 gene, encodes pchA protein). In our genetic circuit, pchA is used to bind the promoter of LEE1, PLEE1, and then help to activate the expression of its downstream CI gene.
