crp_trigger_2_phi29

crp_trigger_3_phi29 (BBa_K4438905) is a single-stranded DNA having 34 nucleotides. Figure 1A) shows the secondary structure and minimum free energy. Its 3’ end has a few bases complementary to the part Aptamer_crp (BBa_K4438900). At the 5’ end, there are few bases of broccoli cDNA. The middle region has an ssDNA antisense T7 promoter sequence.


Usage and Biology

The part crp_trigger_3_phi29 (BBa_K4438905) binds to the part Aptamer_crp (BBa_K4438900) due to complementarity. Figure 1D) Shows the secondary structure of both parts hybridised at 37° Celsius. CRP binds with Aptamer_crp (BBa_K4438900) and displaces the crp_trigger_3_phi29 (BBa_K4438905) [1]. This part has complete complementarity with part crp_Target_3(BBa_K4438906). Phi 29 DNA extension polymerase extends the template strand and in-vitro transcription of the duplex forms multiple broccoli light-up aptamers. Different levels of CRP can be detected using all these three parts.
