MN4_trigger_1-T7promoter-w/oPol

It is a single-stranded DNA sequence having 19 nucleotides (ssDNA sense T7 promoter strand). Figure 1A) shows the secondary structure and minimum free energy.

Usage and Biology

This part can bind to Aptamer_MN4 (BBa_K4438800) due to complementarity and disrupt its three-way junction. Figure 1D) Shows the secondary structure of both parts hybridised at 37° Celsius. Quinine binds with cocaine-binding aptamer MN4 [1] and displaces the MN4_trigger_1-T7promoter-w/oPol(BBa_K4438801). This part has complete complementarity with MN4_Target_1(BBa_K4438802). T7 polymerase binds and in-vitro transcription of the duplex forms multiple broccoli light-up aptamers. All three parts can be used to detect different concentrations of quinine.
