CRISPR array
The sequence includes two repetitive sequences recognisable by the Type-IC Cas proteins. The second repetitive sequence is modified to prevent recombination, so the spacer sequence that is placed in between does not get flipped out. The sequence found in the middle of the repeats is a random DNA sequence, that can be substituted by a spacer targeting a desired locus in the genome. BsaI sites can be found at the end of the first repeat and at the beginning of the second, so a new spacer can be integrated through simple restriction-ligation.
