XOR gate
The XOR gate is a type of Boolean logic gate, which is similar to the OR gate. The difference is that it can only be used to activate downstream gene expression in response to the upstream corresponding promoter in the presence of any marker. When all markers corresponding to the upstream promoter are present at the same time, the XOR gate will be closed.
Usage and Biology
The XOR gate is constructed with a p7 promoter and a terminator ( BBa_B0015 ). We added cis-trans potential Bxb1 integrase and Tp901 integrase sites on both sides of the terminator to form the so-called XOR gate. Finally, we introduced RiboJ and BCD2 sequences to ensure smooth transcription with the XOR gate on.
The XOR gate can be assembled to any biobrick containing an RBS site. You can use the XOR gate to respond to multiple promoters upstream, in the presence of any one or several of the corresponding markers. To demonstrate its efficacy, we designed RFP proteins downstream of it to test it (BBa_K4156118).
Meanwhile, we plan to apply the OR gate on the expression of HlyE (BBa_K4156119). Together with the use of upstream regulatory sequences, we can control the expression of HlyE only when needed, while avoiding any undesired production and harm.
