Quorum sensing suicidal system-part1
what is it
The quorum sensing suicidal system consists of three parts: LUXR, CRISPRi/dCas9, and sgRNA. The LUXR protein is regulated by the T7 promoter, lac operator, and T7 terminator. CRISPRi/dCas9 is regulated by a quorum sensing promoter and quorum sensing terminator. sgRNA is regulated by the H1 promoter and H1 terminator.

At first, if the IPTG is absent, the RNA ligase in the system will converge with the T7 promoter which can not lead to the production of mRNA. Thus, protein and enzyme production will be inhibited. However, in severe situations, IPTG could be present in the natural environment, so we have considered a double safety system. If LUXR detects the presence of AHL, a quorum sensing chemical sent out by E.Coli, quorum sensing promoter will be activated. Later, the promoter will activate the CRISPRi/dCas9 system targeting the ATP ligase of E.coli. Consequently, without ATP, the bacteria will be forced to die.
