Gentamicin Resistance Gene cassette

Introduction: Gentamicin is an aminoglycoside frequently used in clinical settings to treat many bacterial infections. A gentamicin resistance gene can confer resistance to aminoglycosides based on several mechanisms: enzymatic inactivation of aminoglycosides, increased efflux from the bacterial cell, interfering with binding domains of aminoglycosides, etc.

Usage and Biology

This part is the entire circuit of the Gentamicin Resistance gene which is used within the plasmid (pFastBac-YmBac2) and it confers resistance to gentamicin. The expression of the gentamicin coding sequence is driven by the pC promoter BBa_K4235004. This circuit is also a part of the mini-attTn7 transposon segment (add link), used in the Bac-to-Bac expression system by Stony_Brook 2021.

This marker is used to provide gentamicin resistance to the pFastBac vector BBa_K4235002 during the transposition in the Bacmid, propagated in E coli DH10Bac cells. A concentration of 7 ug/ml gentamicin is recommended for transposition.
