DNA template of target RNA for LbuCas13a
This part encodes a target RNA template that serves as a target sequence for Cas13a recognition. Once Cas13a protein binds to sgRNA to form the Cas13a-sgRNA complex, Cas13a in the complex can recognize protospacer flanking sequences (PFS) of target RNA, and then change the conformation of Cas13a by base complementary pairing between sgRNA and target RNA after PFS recognition. Cas13a exhibits RNase activity to cleave specific targeted single-guided RNA (cis-cleavage activity). The cleavage of double-stranded DNA activates the ability of Cas13a protein to cleave non-specific ssDNA (trans cleavage activity).
In general, this is a piece of DNA that is transcribed to produce the target RNA, and we can verify the identification of false positives and other phenomena by synthesizing this target RNA.
It should be noted that this sequence has no potential to encode proteins and is only used to transcribe RNA.
