OPH (organophosphate hydrolase) + his-tag
Our experiment aims to resolve environmental eutrophication, which is caused by the abundance of organic phosphate and nitrogen in the water. OPH, a gene that encodes organophosphate hydrolase, paraoxon, a type of organic phosphate and insecticide. We use paraoxon as the substrate of OPH's degradation, after which it would be converted into diethyl phosphate (DEP) and p-nitrophenol (pNP). We use pNP as an indicator for spectrophotometric identification. his-tag is attached to the end of the OPH gene for the purification of the protein.
