Phytophthora Toehold Switch Sequence

The Phytophthora Toehold Switch Sequence was derived from NUPACK and was constructed with the lowest normalized ensemble defect rate possible. The sequence forms a hairpin loop, hiding machinery necessary for translation. However, in the presence of the X24 gene of Phytophthora cryptogea, a plant pathogen, the hairpin loop unravels, allowing for expression of the downstream gene to occur. As a result, this sequence can be paired with a reporter gene to function as a biosensor for the presence of the plant disease P. cryptogea.
