22 Amino Acid Linker

Flexible 22 amino acid linker (LEGSGQGPGSGQGSGSPGSGQG) that can be used to space out two domains in a fusion protein so that the domains do not interfere with each other and fold independently.

Usage and Biology

The first 15 amino acids were taken from Bisson-Filho et al. (Science, 2017), while the other 7 amino acids consisted of similar repeated motifs as the first 15 amino acids. This long linker was used in our project to space out the OpdA enzyme from the SpyTag002 module in the OpdA-SpyTag002 family of fusion proteins, in order to preserve the folding and hence the activity of the enzyme as well as the isopeptide bond formation ability of the SpyTag002 with SpyCatcher002.
