Coding sequence
In order to improve the quality of SeNPs and improve the reduction efficiency, we hope to link those standard parts and then, two genes express at the same time. This part consist of low intensity SefA and low intensity ssuE. After verifying the function of SefA and ssuE separately, we integrate them and obtain nine combinations of different promoter strengths. And the low SefA and the low ssuE Can produce the best SeNPs.
In order to explore the optimal conditions for the production of SeNPs,we used the Response surface model.We hope to maximize SeNPs production by optimizing temperature, pH and selenite concentration. First, we conducted a preliminary experiment on these three factors, and we chose the best three gradients(Fig.1) of these three factors to do further experiments.
