P28a Promoter
This promoter is activated by sigma factor σ28, which directs RNA polymerase to transcribe the genes required for flagellar biosynthesis and chemotaxis in many bacteria, such as Escherichia coli, Bacillus subtilis and so on. We utilize P28a promoter to express the tetR repressor in a Cell-Free system.
Usage and Biology
In E. coli, the sigma factor family competes for the same core RNA polymerase. σ28 is a specific transcription factor concerning the FliA in the sigma factor family, is the alternate sigma factor for the class 3 flagellar operon. σ28 and its promoter are highly specific, with leakage through promoter P28 almost systematically below the limit of detection. To increase the expression intensity of each component, we used σ28 as a promoter factor.
