Design Notes

This gene was synthesized commercially from the published sequence (IDT). For an initial proof-of-concept, we introduced the binding sites into a commercially available pcDNA5 plasmid (Thermo Fischer, V103320) with mCherry fluorescence protein. HEK cells were transfected with these plasmids and imaged for fluorescent activity.

This is an identified binding site for the DhdR gene, and is generated from sequence analysis. This sequence contains a spacer attached to the end, as literature sources indicated that the presence of a spacer might be able to improve expression activity.
