Usage and Biology

The CRISPR targeting specificity is determined both by base pairing between the RNA chimera and the target DNA and by the binding between the Cas9 protein and a short DNA motif commonly found at the 3' end of the target DNA, called the protospacer adjacent motif (PAM). The N20 sequence is 5’-GGAATAACCGCTTCCCTCCC-3’. This sequence will help the Cas9 protein and gRNA complex to locate the fadR knockout site.
