Design Notes

Hammerhead ribozymes were introduced in order to ensure, that the transcript does not include artifacts from promoter or terminator. Those sequences could interfere with the secondary structure of the RNA. The hammerhead ribozymes were designed with help of the DesignSimple software.

The secondary structure of the RNA was predicted in order to ensure that the linker region does not interfere with the functional entities (hairpins).
