AIP is induced and fluorescence is generated
This part contains agrAC gene, which constitutes the AIP sensing system. By connecting the sfGFP gene downstream, we were able to verify the expression intensity of this part.
Usage and Biology
AgrC protein is the reporter for AIP. Upon binding to AIP, AgrC dimerizes and phosphorylates each other to activate AgrC proteins. AgrC can phosphorylate AgrA protein. The phosphorylated AgrA protein dimerizes and binds to the P2 promoter of the agrBDCA operon to form a positive feedback loop.
Characterization
To verify the construction of agrAC-P2-sfGFP, the digestion and agarose gel electrophoresis were performed by a standard protocol (Fig. 1).
