Design Notes

The actual sequence of crRNA is AAUUUCUACUGUUGUAGAUAUAGAGAAUUGUAACUG. As part of CRISPR-Cas12a system, we adopted the sequence of BBa_K3696008 (crRNA) from other articles. Therefore it is essential to be careful with ssDNA that will bind with crRNA in Cas12a reaction, because these ssDNA may be affected by the design of upstream signal sources.

Source

The sequence of BBa_K3696008 is artificially designed. We adopted the sequence from the article "CRISPR-Cas12a has both cis- and trans- cleavage activities on single-stranded DNA" and synthesized the part accordingly.
