Transcriptional Synchronization Cassette (Eukaryotic Version)
The eukaryote plant cassette consists of mCherry, a poly-A tail, HDVRz, HHRz, sense YFP and antisense YFP. mCherry is acting as reporter. HDVRz inhibits the attachment of a functional poly-A tail, therefore an artificial one was added to the sequence. HHRz is cutting at the 3'-end of the sense YFP and at the 5'-end of the antisense YFP. The connection between sense YFP and antisense YFP is formed by a spacer inheriting a loop, so after the cleavage a double strand is formed. The plant itself possesses a mechanism to destroy the loop, thus breaking the previously formed double strand. The antisense strand can then be used by the RNAi machinery for further silencing processes.
