Usage and Biology
This composite part is used as the serotonin degradation device. It is composed of the basic parts BBa_K3594008, Monoamine Oxidase (MAOA) gene BBa_K1615022 and Aldose reductase (ALDR) gene BBa_K1602004.
Characterization
According to the testing result of SDS-PAGE, although we have successfully constructed the plasmid, MAO is not expressed by the bacteria. Therefore, we used mathematical modelling to predict the concentration of MAO and other following effect. We assume that the change of protein concentration in the external environment is related to the amount of gene expression, the rate of bacterial lysis, the rate of protein degradation, and the rate of locust intestinal peristalsis.
And got the following results:
The ordinate represents the protein concentration. It can be seen from the image that the protein increases rapidly after the bacterial self-lysis. After reaching a certain concentration, due to the degradation rate of the protein itself and the locust intestinal peristalsis. The rate decreases slightly, and the concentration increases with the next bacterial self-lysis, which forms a stable cycle that can release enzymes efficiently.
The abscissa represents time, and the ordinate is the concentration of serotonin in the locust. It can be seen from the image that the concentration of serotonin decline over time. Sequence and Features
