Summary

Linkers are generally composed of small, non-polar or polar amino acids as suggested by Argos. The small size of these amino acids provides flexibility, and allows for mobility of the connecting functional domains. Flexible linkers are generally rich in small or polar amino acids such as Gly and Ser to provide good flexibility and solubility. They are suitable choices when certain movements or interactions are required for fusion protein domains. In addition, the length of the linkers can be adjusted to allow for proper folding or to achieve optimal biological activity of the fusion proteins.
