Constitutive RFP Promoter in E. coli
The promoter sequence acts as a repressive promoter in the presence of a lux R cassette (system that works between lux R protein and N-3-oxohexanoyl-L-homoserine lactone (AHL) to repress the expression of a lux gene), but because the lux gene is not expressed in E. coli is there is no lux R protein present, the promoter will likely be constitutive. This means that the gene upstream of the promoter will always be transcribed. Sticky ends have been added to the oligos so that they can be inserted into plasmids, which by the process of bacterial transformation will replicate and allow progeny to express the gene upstream of the promoter. The promoter should cause the E. coli colonies to be express RFP.
