Design Notes

The OxyR protein transcription factor was designed using principles relating to statistical data of structural features of reactive cysteines in a wide variety of proteins. Constitutive promoter was chosen for the production of the OxyR protein in order to reach a high enough level to be able to bind with H2O2 and induce the downstream genes controlled by TrxC promoter.


Source

This composite part was made from largely Existing BioBricks. Constitutive promoter, RBS and Terminator sequences are from the commonly used parts on the registry. The inducible promoter TrxCp was designed by the Yang Ming University iGEM team 2013. While the part BBa_K3031018 is one of our new basic parts for the 2019 season - it is an OxyR(mutated) gene sequence which codes for the transcription factor (TF) OxyR protein.Detailed overview of what we did to decide upon mutations to the coding sequence can be found on our Wiki Page for model. OxyR amino acid sequence was obtained from Uniprot and then the nucleotide sequence was codon optimized by Genscript.
