Theophylline actif aptazyme

An aptazyme is an aptamer-ribozyme combination with intrinsic RNA cleavage activity. This aptazyme sequence is specific for the theophylline (ligand). Theophylline (1,3-dimethylxanthine) is a drug used in therapy for respiratory diseases and is structurally similar to theobromine and caffeine. Aptazyme is composed of 3 different parts: a twister ribozyme module, a communication module and an aptamer module.

Figure 1: Theophylline aptazyme secondary structure. The Shine-Dalgarno (SD) and the anti-SD are shown in light blue and grey, respectively. The inactivating mutation is highlighted in yellow. The cleavage site is indicated with a grey arrow. The figure is reproduced from [Stifel, J., Spöring, M., and Hartig, J.S. (2019). The described anti-theophylline aptazyme is an switch-off. Its catalytic activity (self-cleavage) requires the absence of ligand.
