L7Ae (Transcriptional level repressor)

L7Ae is a transcriptional level repressor which targets RNA K-turns.



2019 SYSU-CHINA's characterization

SYSU-CHINA added some quantitative experimental characterization data on the L7Ae protein.
L7Ae is a protein targets RNA K-turn. We characterizated L7Ae and constructed an L7Ae mutation(L7AeO,BBa_3179001) to study the suppression efficiency of the L7Ae-Kturn System.

The result shows that the mutation couldn’t work normally but still functions in high concentration. Meanwhile, the 1000ng/ml L7Ae can depress the translation of mRNA with a efficiency of 84%.

Now the sequence analysis and computer modeling have shown the predicted structure of L7Ae. We can easily see that there are some α-helixes in L7Ae, and one of them is the bidding site.

Cytotoxicity test

SYSU-CHINA tested the cytotoxicity of L7Ae, which was previously considered to be slightly cytotoxic, but the opposite result was found in our experiments.Neither L7Ae nor its mutants showed significant cytotoxicity.
