Biology
A Ribosome Binding Site (or RBS) is a short DNA sequence present upstream of the transcriptional unit. In prokaryotic systems, it is easily identified by the presence of a Shine-Dalgarno like sequence which pairs with the anti-Shine-Dalgarno like sequence of the 3' end of 16S rRNA. An RBS is crucial for the process of translation as it dictates the binding of the ribosome (the translational machinery) to the mRNA transcript encoding the protein of interest.
The effectiveness (or strength) of the RBS sequence depends on the base-pairing potential of the RBS to the 16S rRNA alon with the spacing between the sequence and the start codon. sRBS happens to be a strong ribosome binding site enhancing translational efficiency.
Usage
The sRBS sequence can be used upstream of any gene whose product is desired to be enhanced. This part is used to maximise the translational efficiency of the CcaR protein in the composite gene constructs.
