Spider silk serves as a new material with superior properties that can be applied in medication, cloth, and aerospace fields. However, spider breeding is not applicable due to spider's fierce behavior. The current approach is to produce recombinant spidroins (silk proteins) from other chassis and spin them into silk. NT minispidroin composed of an NT from E. australis MaSp1(major ambulate spidorins) . The N-terminal domain is a non-repetitive and monomeric protein that has a high aqueous solubility in neutral pH solution, and the entire spidorin’s high solubility in pH=7 solution is derived from the NT. Due to the pH responsiveness of the NT, it will form stable dimers that can lock the entire spidorin into large networks as pH of the solution in the spinning duct decreases. 2Rep is the important repetitive region that is responsible for the beta-sheet corroboration when pH decrease. We here use amilCP combined to the end of the extensive repetitive region-2Rep, in order to produce recombinant chromoprotein that have higher solubility due to the presence of pH.
