Design Notes

Here we have put a lldRO1-J23117-lldRO2 along with B0034 (which is a strong RBs) and downstream to it, we have attached YebF-IL12 and FLAG-tag.

Source

YebF-IL12-FLAG tag was synthesized by Twist Bioscience. lldRO1-J23117-lldRO2 + B0034 was obtained as gBlock from IDT and these were joined together using SOEing (Spliced Overlap Extenstion) PCR.
