8*suntag

Signals in many biological processes can be amplified by recruiting multiple copies of regulatory proteins to a site of action. Harnessing this principle, a novel protein scaffold, a repeating peptide array termed SunTag was developed, which can recruit multiple copies of an antibody-fusion protein. Suntag comes from an antibody-peptide pair linked with GCN4, which significantly amplifies the glowing signals of green fluorescent proteins. Suntag is essentially a set of molecular hooks that hook multiple copies of biologically active molecules onto protein scaffolds that target to some genes or molecules. Compared with the assembled molecules without these hooks, the biological activity of the molecules integrated with SunTag was significantly amplified. Here we hope that IL-15 linked with suntag can better mediated receptor dimerization. SunTag8×consists of a string of eight GCN4 peptide tags. Moreover, to determine the best amount of these peptide tags, we set different enzyme cutting sites between every 2 GCN4 single chains, after cutting with different restriction enzymes, we got the other three types of IL-15 proteins labeled with different lengths of suntags: IL-15-suntag6×, IL-15-suntag4×, IL-15-suntag2×.
